ABSTRACT

OBJECTIVE:To detect curative mechanism of bushen huoxue huatan formula on polycystic ovarian syndrome (PCOS)model rats. METHODS:50 rats were randomly divided into normal control group,model control group,positive control group [rosiglitazone,3.0 mg/(kg·d)] and Bushen huoxue huatan formula high-dose,low-dose groups [7.5,5.0 g/(kg·d)],10 in each group. Except for normal control group,the PCOS model was induced by Poresky method in other groups. After modeling, normal control group and model control group were received normal saline intragastrically,medicine groups were received corre-sponding liquids intragastrically,once a day,for 22 d. After administration,the serum levels of testosterone (T),anti-mullerian hormone (AMH),insulin-like growth factor Ⅰ(IGF-Ⅰ),tumor necrosis factor-α(TNF-α) and the positive expression of trans-forming growth factor-β1(TGF-β1)in ovarian tissue were determined. RESULTS:Compared with normal control group,T,AMH, IGF-Ⅰ,TNF-αlevels in serum and TGF-β1 positive expression rate in ovarian tissue of rats in model control group were significant-ly increased(P<0.05);compared with model control group,T,AMH,IGF-Ⅰ,TNF-α levels in serum and TGF-β1 positive ex-pression rate of rats in ovarian tissue in other medicine groups were significantly decreased(P<0.05),and the indexes in Bushen huoxue huatan formula different doses groups were superior to positive control group(P<0.05). CONCLUSIONS:Bushen huoxue huatan formula can obviously reduce the T,AMH,IGF-Ⅰ,TNF-α levels in serum and TGF-β1 positive expression rate in ovarian tissue in model rats with PCOS,which may be one of the mechanisms of its treatment for PCOS.

ABSTRACT

OBJECTIVE:To explore the effects of Bushen huoxue huatan decoction on ovarian blood supply and form in model rats with polycystic ovarian syndrome(PCOS). METHODS:50 SD female rats were randomized into normal control group,model group,positive control group [rosiglitazone,3 mg/(kg·d)],Bushen huoxue huatan decoction low-dose and high-dose groups [5.0, 7.5 g/(kg·d)],with 10 rats in each group. Except for normal control group,PCOS model was induced in the other groups by Poresky method. After modeling,treatment groups were given relevant medicine intragastrically. The normal control group and mod-el group were given normal saline [10 mL/(kg·d)] intragastrically,once a day,for 22 days. After the end of administration,the rats were sacrificed. The microvessel density (MVD) was recorded. mRNA expression of vascular endothelial growth factor (VEGF), ovarian angiotensin Ⅱ receptor(Ang Ⅱ)type 1(AT1)and type 2(AT2),G-protein coupled receptor MAS were all detected in ovarian tissue of rats. The ovarian form was observed. RESULTS:Compared with normal control group,MVD of mod-el group and treatment groups were all increased,while mRNA expression of AT1,AT2 and MAS were down-regulated (P<0.05);cystic sinus sollicle,the decrease of granular cell layer,egg mother cells disappearance,the decrease of luteal tissue and other obvious pathological changes could be found in ovarian tissue of rats in model group. Compared with model group,above in-dexes of treatment groups were improved significantly,and the effects of Bushen huoxue huatan decoction low-dose and high-dose groups were better than that of positive control group (P<0.05). The pathological changes of ovarian tissue in treatment groups were all improved to certain extent. CONCLUSIONS:Bushen huoxue huatan decoction can improve ovarian blood supply,angio-genesis and ovulation in PCOS rats. It is beneficial to improve ovarian form.

ABSTRACT

Objective@#To evaluate the feasibility of combining simultaneous triple semicircular canal plugging (TSCP) and cochlear implantation (CI) to treat vertigo and hearing loss in advanced Meniere's disease(MD) patients, so as to provide an alternative surgical procedure for treating this disorder.@*Methods@#Data from seven patients, who were referred to our hospital and diagnosed with unilateral MD strictly meeting the criteria issued by Chinese Academy of Otolaryngology-Head and Neck Surgery Committee (2006), from Jan. 2015 to Jan. 2016, were retrospectively analyzed in this work. Seven patients, in whom the standardized conservative treatment had been given for at least one year and frequent vertigo still occurred, underwent simultaneous TSCP and CI under general anesthesia via mastoid approach. Postoperative follow-up time was more than six months. Vertigo control and auditory function were measured. Pure tone audiometry, speech perception scores, caloric test, head impulse test (HIT), and vestibular evoked myogenic potential (VEMP) were performed for evaluation of audiological and vestibular functions.@*Results@#All patients had bilateral severe sensorineural hearing loss preoperatively. One side hearing loss was due to MD and another side was due to reasons including sudden sensorineural hearing loss, mumps and other unknown reason. The total control rate of vertigo in seven MD patients was 100.0% in the six-month follow-up, with complete control rate of 85.7% (6/7) and substantial control rate of 14.3% (1/7). Improved hearing threshold and speech perception scores were observed in all study participants. Postoperative average aided hearing threshold was 32.5 dBHL, the average monosyllabic word score was 42.6% and speech perception scores of sentences tested in quiet was 52.3%. Tinnitus improved in five cases, and no significant change in two patients. Post-operatively, all patients suffered from temporary vertigo and balance disorders. Vertigo was disappeared in all patients within 3-5 days, while, an average recovery time of balance disorders was 19.7 days. Six months after treatment, loss of semicircular canal function by caloric test was found in the operation side of all patients and no change in cVEMP or oVEMP test was noted. All patients had no facial paralysis, cerebrospinal fluid leakage, and other complications.@*Conclusions@#A combined approach of TSCP and CI which could control vertigo effectively and improve hearing loss and tinnitus represents an effective and safe therapy for some advanced MD patients.

ABSTRACT

OBJECTIVE:To study the effects of Tiaojing zhuyun capsules on the number of blastocyst implantation,estrogen, progestogen and their receptors(ER,PR)in mice with implantation dysfunction. METHODS:48 pregnant mice were randomly di-vided into normal control group (normal saline),pathological model group (normal saline) and Tiaojing zhuyun capsules group (24 g/kg),with 16 mice in each group. They were given relevant medicine intragastrically,once a day,for consecutive 3 d. On the 4th day,those mice were given mifepristone 0.1 ml subcutaneously to induce implantation dysfunction model except for normal control group. On the 5th day,the mice were sacrificed. The number of blastocyst implantation sites was observed. The serum lev-els of estradiol (E2) and progesterone (P) were determined by radioimmunoassay,and the expression of ER and PR protein were determined by immunohistochemistry. RESULTS:On the 5th day of pregnancy,the number of embryo implantation sites,serum levels of E2 and P and the protein expression of ER and PR in pathological model group were significantly lower than in normal con-trol group(P<0.05);the number of embryo implantation sites,levels of E2 and P,the protein expression of ER and PR in Tiao-jing zhuyun capsules group were significantly higher than in pathological model group (P<0.05). CONCLUSIONS:Tiaojing zhuyun capsules can effectively improve the serum levels of E2 and P,the protein expression of ER and PR,and increase the num-ber of blastocyst implantation sites in mice.

ABSTRACT

OBJECTIVE:To investigate the effects of Tiaojingzhuyun capsules on endometrial receptivity in mice with implanta-tion dysfunction and corresponding mechanism. METHODS:Pregnant mice were randomized into normal group,model group and the groups of low and high-dose [12,24 g/(kg·d)] Tiaojingzhuyun capsules,with 10 mice in each group. The mice in the drug ad-ministration groups were given corresponding drugs intragastrically,while those in the normal group and the model group were giv-en normal saline intragastrically,for 3 consecutive days from the 1st day of pregnancy. On the 4th day of pregnancy,the mice in all groups except for the normal group were given mifepristone solution subactaneously to establish model of implantation dysfunc-tion. On the 5th day of pregnancy,the mice were sacrificed,and then the change in endometrial morphology was observed and en-dometrial maturity was scored;the protein expression levels of leukaemia inhibitory factor(LIF),homoeobox gene HOXA10,ma-trix metalloproteinase 9(MMP-9)and matrix metalloproteinase inhibitory factor 1(TIMP-1)expression in endometrial tissues were determined. RESULTS:Compared to the normal group,the mice in the model group and the drug administration groups had endo-metrial hypoplasia and obviously lower endometrial maturity score and protein expression levels of LIF,HOXA10,MMP-9 and TIMP-1 in endometrial tissues(P<0.05). Compared to the model group,the mice in the drug administration groups demonstrated endometrial development which improved or tended to be normal and had significantly higher indexes mentioned above(P<0.05);besides,the effect shown in the group of high-dose Tiaojingzhuyun capsules was stronger than that in the group of low-dose Tiao-jingzhuyun capsules (P<0.05). CONCLUSIONS:Tiaojingzhuyun capsules can improve the endometrial receptivity in mice with implantation dysfunction by a mechanism which may be related to upregulating the gene expression of HOXA10 and LIF,and main-taining the balance of MMP-9 and TIMP-1.

ABSTRACT

To investigate roles of nitric oxide (NO) signal in accumulations of phenolic acids in abscisic.acid (ABA)-induced Salvia miltiorrhiza hairy roots, S. miltiorrhiza hairy roots were treated with different concentrations of sodium nitroprusside (SNP)-an exogenous NO donor, for 6 days, and contents of phenolic acids in the hairy roots are determined. Then with treatment of ABA and NO scavenger (2-(4-carboxy-2-phenyl)-4,4,5,5-tetramethylimidazoline-1- oxyl-3-oxide, c-PTIO) or NO synthase inhibitor (NG-nitro-L-arginine methyl ester, L-NAME), contents of phenolic acids and expression levels of three key genes involved in phenolic acids biosynthesis were detected. Phenolic acids production in S. miltiorrhiza hairy roots was most significantly improved by 100 µmoL/L SNP. Contents of RA and salvianolic acid B increased by 3 and 4 folds. ABA significantly improved transcript levels of PAL (phenylalanine ammonia lyase), TAT (tyrosine aminotransferase) and RAS (rosmarinic acid synthase), and increased phenolic acids accumulations. However, with treatments of ABA+c-PTIO or ABA+L-NAME, accumulations of phenolic acids and expression levels of the three key genes were significantly inhibited. Both NO and ABA can increase accumulations of phenolic acids in S. miltiorrhiza hairy roots. NO signal probably mediates the ABA-induced phenolic acids production.
